Does Access-to-MySQL require ODBC drivers?

MS Access to MySQL converter does not require ODBC drivers. It requires Data Access Objects (DAO) library produced by Microsoft to read the source database. Since some versions of MS Access/Office do not include required DAO DLLs, you may need to install Microsoft Access Runtime:

Despite of the fact you have 32- or 64-bit version of OS and MS Office, it is necessary to download and install 32-bit version of Access Runtime (the corresponding file name ends with "x86_en-us.exe").
